چکیده

Viper’s venom is a source of biopharmaceutical compounds, hence the need to assess the effect of this animal extract on human blood. Here, we studied the blood coagulation disorders and hemolytic activities of the venom of M. bornmulleri viper. The pro-coagulant and anticoagulant effects are analyzed with venom concentrations ranging from 0.4 to 0.0031 mg/mL. Thus, the PT is way above the normal value indicating an anticoagulant activity whereas for the aPTT, the high concentration of the venom showed an anticoagulant activity, but a pro-coagulant effect was occurred when the venom concentration decreases to 0.05 and/or 0.025 mg/mL. Hemolytic tests, performed in suspension (30% RBCs) and on blood agar plate (5% RBCs), show that an increased concentration of the venom going until 1.6 mg cannot produce a hemolytic effect, even in the presence of Ca 2+ (hemolysis < 0.5%). Also, on the blood agar plate no hemolytic area appeared even with 0.04 mg of the lyophilized venom. Otherwise, the venom was able to induce a low hemolytic activity (hemolysis = 1.3 %) by acting on L-α-PC used as substrate. In this case, the destruction of erythrocytes increased proportionally to the added amount of phospholipids which are hydrolyzed to fatty acids and lysophospholipids (two toxic substances for RBCs), probably due to the presence of PLA2 in the venom and which are known by their ability to hydrolyze lecithin Open Journal of Hematology

The relaxant effect of the Montivipera bornmuelleri snake venom on vascular contractility

Molecular richness of snake venoms is an important source of proteins and toxins with potent effects on the cardiovascular system. The alteration of the vascular system in the victim after a venomous snake bite is usually expressed by a significant decrease in blood pressure. Therefore, exploring snake venom to extract and characterize its biomolecules is of considerable medical interest, and f...

Evidence for in vitro antiophidian properties of aqueous buds extract of Eucalyptus against Montivipera bornmuelleri venom

Because snake venoms are complex mixtures of bioactive molecules, snake bites produce a large panel of symptoms which cannot be totally prevented by current antivenoms. Thus investigating plant extracts for antivenomics therapy approaches seemed relevant. Here, we evaluated the potency of the aqueous Buds extract of Eucalyptus (ABEE) to counteract the main enzymatic activities of Montivipera bo...

Blood Coagulation Induced by Iranian Saw-Scaled Viper (Echis Carinatus) Venom: Identification, Purification and Characterization of a Prothrombin Activator

Objective(s): Echis carinatus is one of the venomous snakes in Iran. The venom of Iranian Echis carinatus is a rich source of protein with various factors affecting the plasma protein and blood coagulation factor. Some of these proteins exhibit types of enzymatic activities. However, other items are proteins with no enzymatic activity.   Materials and Methods: In order to study the mechanism ...

Effect of α-lipoic Acid on Hemolytic Activity of Iranian Vipera Lebetina Venom

Abstract Background and Objective: Snake venom is a complex of several toxic elements and enzymes. It has the agents with the ability to destroy cellular and subcellular membrane and to bring about hemolysis of red blood cells (RBC). Two types of direct and indirect hemolytic activity are known in snake venom in that phospholipase A2 is responsible for the indirect lysis. The aim of this st...

Evaluation of Iranian Snake ‘Macrovipera lebetina’ Venom Cytotoxicity in Kidney Cell Line HEK-293

Background:Envenomation by Macrovipera lebetina (M. lebetina) is characterized by prominent local tissue damage, hemorrhage, abnormalities in the blood coagulation system, necrosis, and edema. However, the main cause of death after a bite by M. lebetina has been attributed to acute renal failure (ARF). It is unclear whether the venom components have a direct or indirect action in causing ARF. T...
